Overview:
LJ100™ is a
standardized Eurycoma Longifolia extract, containing 22% Eurypeptides, a
bioactive glyco protein that is now clinically proven for its androgenic
properties. The plant is also commonly known as Tongkat Ali (Ali’s
walking stick) and is commonly found along the hilly jungle slopes of Malaysia.
The name Ali is associated with a male warrior who was well known for his
prowess. Besides its aphrodisiac properties, Eurycoma has been used as
traditional remedies for fever, ulcer, malaria, reducing high blood pressure and
fatigue.
LJ100™ has undergone a patented BAT
extraction process to capture 22% biologically active Eurypeptides™,
proven in research to be responsible for increasing libido, improving sports
performance, increasing fertility, and activating the enzymes that metabolize
the various androgens. Created by the original researchers at the Forest
Research Institute of Malaysia (FRIM) and University of Malaya, LJ100™
(22% Bioactive Eurypeptides) has shown - in human clinical trials - an ability
to increase DHEA and free testosterone, decrease Sex Hormone Binding Globulin
(SHBG), improve HDL, modulate cortisol, and increase IGF-1 level.
The Effects of LJ100TM on Fertility &
Sexual Behavior in Mice:
Between 1993 and 1994, Dr. Johari Saad, head of the
Biochemistry Department at University of Malaya conducted multiple mice studies
to evaluate the fertility and aphrodisiac effects of LJ100™. 48 mice were
divided into 3 sets of 4 groups. Group 1 was treated with saline; Group 2 female
was treated with LJ100™ and male with saline; Group 3 both male and
female treated with LJ100™; Group 4 male treated with LJ100™ and
female with saline. In this study, we observed that the treated males were very
aggressive sexually and showed a higher frequency of mounting compared to the
control group.

Group 3 and 4, where male mice were treated with
LJ100™, yielded a litter size up to 76% larger than the control
group. The increase was due to increased sperm concentration (125% increase),
motility (38% increase), and velocity which increased fertilization rates (82.7%
vs. 94.8%). Group 3 and 4 also showed a 3:1 male to female offspring ratio.
LJ100™ may have a selective action towards the Y sperm during
spermatogenesis or prefer to enhance the development and fertilization of Y
sperm. Dr. Johari also observed 60% increased energy production through
glycolysis and oxidative phosphorylation, 80% increased ATP production observed
in liver homogenates, 60% increased Cyclic GMP and Cyclic AMP in penile tissues.
Aphrodisiac Properties of LJ100™ (22%
Bioactive Eurypeptides):
Sexual stimulus in males leads to a release of nitric oxide
(NO) into the blood vessels of the corpus cavernosa (erectile tissue). NO
activates enzyme guanilate cyclase activity which increases cGMP (cyclic
guanosine monophosphate ) concentration, which allows for the relaxation of
penile arteries, leading to increased blood flow and penile erection. In the
absence of NO release, cGMP is terminated by enzyme PDE5. cAMP also
affects penile erection. cAMP (cyclic adenosine monophosphate) is
modulated by adenyl cyclase, which result in a decline in the free calcium
concentrations, which allows for the relaxation of penile smooth muscle leading
to vasodilatation and erection.
In 2002, Dr. Azimahtol, Faculty of Science & Technology from
the University Kebangsaan Malaysia conducted a study to evaluate the effect of
LJ100™ (22% Bioactive Eurypeptides) was compared with the effectiveness
of sildenafil citrate (Viagra) in triggering penile erection. Sildenafil citrate
is a potent PDE5 inhibitor. By blocking the action of PDE5, the NO-induced
increase of cGMP concentration is augmented and prolonged.
Rabbit corpus cavernosum tissues were treated with LJ100™ (22% Bioactive
Eurypeptides) and sildenafil citrate at different concentrations (0, 1.25,
1.875, 2.5, 3.125 and 3.75 µg/ml) and incubation time; cGMP and cAMP levels were
measured. LJ100™ increased the level of cGMP in rabbit corpus cavernosum
to almost 4-fold at 3.125 µg/ml concentration. Sildenafil citrate increases cGMP
in rabbit corpus cavernosum to 5-fold at 10-4M concentration. LJ100™ was
found to increase cAMP levels in corpus cavernosum, even in the absence of
sexual stimulus, and there were no significant increases of cAMP levels in the
corpus cavernosum tissue treated with Viagra. Results of this study validate the
aphrodisiac properties of LJ100™ which increases and enhances the levels
of cGMP and cAMP, which ultimately leads to penile erection.
Androgen Biosynthesis of LJ100™
Testosterone, or male sex hormone, plays a key role in
developing and maintaining masculine sexual organ, and promotes secondary sexual
characteristics, including the appearance of facial hair, sexual desire, and
sexual behavior. Testosterone also stimulates metabolism, which
promote fat burning, increases the formation of red blood cell, and accelerates
muscle growth. Testosterone is synthesized by an enzymatic sequence of steps,
breaking down cholesterol, via activation of CYP17 (17 a-hyroxylase/17,20 lyase)
enzyme to pregnenolone. CYP 17 is involves in the early stage of steroid
biosynthesis. Pregnenolone is the ultimate parent steroid compound.
Pregnenolone is a steroid precursor, manufactured from cholesterol in the body.
Pregnenolone is a precursor to cortisol, DHEA, Progesterone, and Testosterone.

Between 2002 and 2003, a study was conducted by Dr. Harniza
and Dr. Johari from the University of Malaya to isolate the bioactive
Eurypeptides and test the biosynthesis of various androgens. The in vitro
results from this study showed that Eurypeptides significantly increased CYP17
enzymes compared to the positive control, suggesting that more of the CYP17
enzyme is being produced to metabolize pregnenolone to yield more testosterone
and progesterone. We have also seen 180% increase in testosterone and 190%
increase in progesterone.

Low amounts of pregnenolone and 17-OH pregnenolone compared to
control also confirm that more CYP17 is activated to convert pregnenolone and
17-OH pregnenolone into progesterone and testosterone. Pregnenolone is also a
precursor for the synthesis of pheromones. The study showed that Eurypeptides
influence the synthesis of pheromones by significantly increasing An
a--a pheromone that plays an important role in communication,
psychological and sexual behavior in humans, confirming the aphrodisiac aspects
of
LJ100™.
LJ100™ Natural
Energizer for Healthy Aging of Men:
Testosterone levels drop by about 10 percent every decade
starting at age 30. At the same time, Sex Hormone Binding Globulin (SHBG) is
increasing. SHBG traps much of the testosterone that is still circulating and
makes it unavailable to exert its effects in the body's tissues. Low
bioavailable testosterone levels may cause low sex drive, decreased muscle mass
and strength, increased body fat, osteoporosis, and increased cardiovascular
risk.
In 2003, Dr. Ismail Tambi and Dr. Johari Saad conducted a
pilot human clinical trial at the Specialist Reproductive Research Center,
National Population & Family Development Board, Malaysia. In this study 23
volunteers took 100mg of LJ100™ for three weeks. The Sexual Health
Inventory Questionnaire (SHIQ) results showed 62% of volunteers having an
increased or maximum score, showing increase in sexual desire and success in the
attempts at sexual intercourse. Aging Male Score (AMS) were lowered
demonstrating 91% improvement of sexual score, 73% improvement in the physical
score, 91% improvement of the psychological score, and 52% improvement in
vasomotor score.
Total testosterone levels were not significantly different suggesting that
LJ100™ does not have any effect on steroidogensis. Analysis of DHEA showed
gradual increase from 26% after 1 week to 47% after 3 weeks. This suggests that
LJ100™ may influence the DHEA production, which would in turn be
aromatized to testosterone. SHBG levels were reduced in 36% of the volunteers
after one week and 66% after 3 weeks. This suggests that the extract could have
an effect on the production of SHBG. Consequently, when SHBG level declines, the
Free Testosterone Index (FTI) goes up. FTI showed an escalation of 39% of
subjects after 1 week and 73% after 3 weeks. This study showed that LJ100™
can provide sufficient free testosterone for the body and can be a valuable
health elixir for the aging men.
Anabolic Effects of LJ100™ (22% Bioactive
Eurypeptides)
In a double blind, placebo controlled study conducted by Dr.
Sareena from the Department of Sport Science at the University of Malaya in
2000, 14 healthy adult males received either 100 mg/day of LJ100™ (22%
Bioactive Eurypeptides) or placebo for 8 weeks. Both groups performed an
intensive strength training program for 8 weeks. Although the testosterone level
was not measured during the test period, an increase in fat free mass in
LJ100™ group may be linked to the rise in steroidal hormones in the body.
LJ100™ group showed greater decreased fat mass compared to the placebo; this
could be explained by the higher metabolic rate after consuming LJ100™.
In this study, we observed a greater increment in muscle strength test for the
LJ100™ group which related to a lower muscular neural firing rate, which
we suggest was due to further increased testosterone levels. An increase in arm
circumference observed in the treatment group could be explained by the
testosterone enhancing effect of LJ100™. In conclusion, results obtained
from this pilot study suggest that the administration of LJ100™ (22%
Bioactive Eurypeptides) increased fat free mass, reduced fat mass, increased
muscle strength and size. During and after the administration of LJ100™
there were no adverse effects noted within the LJ100™ group.

Saliva Testosterone Test:
Nine volunteers ages 26-52 were given 200mg of LJ100™
for 10 days. Volunteers 1-5 who engaged in physical exercise experienced a 70%
to 133% increase in Free Testosterone. Volunteers 6-9 who did not exercise
experienced a 35% to 70% increase in Free Testosterone level. This study shows
that exercise will enhance the effects of LJ100™ as it
modulates testosterone level according to the body’s needs.

Human Toxicology and Clinical Study:
In 2004, Dr. Ismail Tambi conducted a double-blind placebo
controlled human clinical trial at the Specialist Reproductive Research Center,
National Population & Family Development Board, Malaysia. 20 male
volunteers of various health conditions from the ages of 38 to 58 were randomly
given either 200, 400, or 600 mg of LJ100™ (22% Bioactive
Eurypeptides) or placebo for 2 months. The SHIM (Sexual Health Inventory for
Male) scores were elevated to upper normal level showing improvement in sexual
desire and performance for a majority of the volunteers. The Aging Male Score
(AMS) were lowered showing improvement in sexual, physical, psychology, and
vasomotor domain for majority of the volunteers. Testosterone and DHEAS levels
showed high normal levels when compared to baseline and is consistent with the
excellent QOL (Quality of Life) score. LJ100™ modulates the
production of DHEAS, which would in turn be aromatized to testosterone.
Bioavailable testosterone is needed to maintain wellness and enhance libido.
Full blood examinations showed normal limits even at the
600mg dosage for all tests. LJ100™ does not interfere with the
platelets or hemoglobin and does not have any toxicity to the RBC. The liver
function test showed no liver toxicity even at high doses of LJ100™.
Renal function test showed that
LJ100™ has no harmful effect on the renal system even for men
whose renal function is affected (as seen in diabetics). Some volunteers even
showed improved or normal levels of uric acid at the end of the trials. Tumor
markers are sensitive indicators of cellular stress. AFP (Alpha Fetal Protein)
identifies testicular cancer; PSA (Prostate Specific Antigen) is used to detect
prostate cancer. All tumor markers were within normal levels even at 600mg
dosage of LJ100™. The lipid studies showed increment of
high-density lipoprotein (HDL Cholesterol –good cholesterol) levels. Elevations
of HDL are normally associated with a lower risk of heart attack. Diabetic
volunteers showed an improved level of blood glucose levels. LJ100™
does not exert hypoglycemic effects but may have an influence on the cellular
uptake of glucose.
The cortisol levels were in the higher than normal level for
the
LJ100™ group as opposed to the placebo group. The positive
benefits of high cortisol are enhancement of immune response during illness and
body defense during health crisis. The negative aspect of cortisol is associated
with the metabolic syndrome X, which includes obsesity, type-2 diabetes,
hypertension, and increased cardiovascular risk. This study suggests that
LJ100™ has influence over the adrenal gland and modulates the
release of cortisol.
Low thyroxin indicates low BMR (Basal Body Metabolism Rate).
The majority of volunteers on LJ100™ have higher than normal
levels of thyroxine compared to the placebo group, meaning higher metabolism
rate which can be translated to weight management.
IGF-1 regulates cellular growth and development. Premature
reduction in IGF-1 causes cell death. IGF-1 is the anti-aging hormone as it
increases muscle bulk and lean body mass and cell regeneration. Volunteers
taking the LJ100™ had higher than normal levels of IGF-1 at
the end of the trials. Some with a low IGF-1 level increased to a normal level
after 1 week. The placebo group showed no change in IGF-1 levels. This study
suggests that LJ100™ has a secretagogue effect on growth
hormone and modulates the release of IGF-1; in turn, making it a great
anti-aging supplement.
Conclusion:
It is concluded through clinical trials and studies that
LJ100™ (22% Bioactive Eurypeptides) is a great overall-health
supplement. It has many positive effects including enhancing libido, improving
sports performance, increasing fertility, activating enzymatic activities of
various androgen hormones, and is a growth hormone secretagogue.
